A novel adaxialized mutant of canola.

Canola (Brassica napus) is an important oilseed crop that is currently the subject of a reverse genetics TILLING screen by the Canadian Tilling Facility (CAN-TILL). An intriguing phenotype was observed in a canola plant grown from seed mutagenized for the CAN-TILL project. This mutant, named LIP (lamina epiphylla), has been characterized using morphological and anatomical methods. It behaves as a single locus and is semi-dominant. Plants with a weak phenotype are predicted to be heterozygous, while homozygotes have variable leaf morphology including strikingly trumpet-shaped leaves often with many filamentous processes, and ectopic epilaminar leaf outgrowths. Anatomically, the leaf petiole of putative homozygous plants is adaxialized, including the development of amphivasal vascular bundles. This radialization of bundles translates into a notably abnormal vascular structure primarily seen in the petiole. Although generally this mutant phenotype is similar to the previously described semi-dominant mutations in the PHABULOSA, PHAVOLUTA and REVOLUTA genes of Arabidopsis thaliana, there are characteristics manifested by this B. napus LIP mutant that cannot be easily observed in A. thaliana (primarily extensive stem secondary thickening, vegetative internode elongation, and leaf lobation in B. napus). This mutant also has floral phenotypes similar to its vegetative structures.
