Genomics / Proteomics Timme, Ruth E. [1], Delwiche, Charles [2]. EST survey of the Charophytes. There must have been many genetic and/or regulatory changes that allowed embryophytes to diversify on land from their freshwater, green algal ancestors. The goal of this project is to uncover these genetic changes by surveying expressed sequence tags (ESTs) from all seven orders of Charophyte green algae (ancestors of land plants). While still in the data collection stage, the current focus is to introduce the general study system, to present both specific hypotheses and a general computational approach to addressing the objectives of this study, and to reveal some preliminary data. Our completed data set will consist of about 180,000 ESTs from seven charophyte green algal species. The first approach will be to identify homologs of land-plant genes in the green algal ESTs that have potential importance in the colonization and/or adaptation to land (i.e. genes involved in life history regulation, cell-differentiation, lignification, association with mycorrhizae, etc.). The second approach will be to computationally identify any gene or region of a gene that has interesting characteristics from an evolutionary prospective within the EST databases, which might include regions under positive or purifying selection, gene duplications, or other unforeseen discoveries. Most of the EST libraries are in the sequencing stage, but preliminary data from Nitella hyalina ESTs enabled us get a first gimpse at identifying interesting land plant homologes in the green algal ancestors.
