Quantitative investigation with GFP expressing plants.

Red-shifted Green Fluorescent Protein (sGFP) has become a useful tool in investigating both plant development and gene regulation. With the addition of an epifluorescent camera attachment illuminated with blue light emitting diode arrays, undergraduates can capture quantitative digital images of sGFP fluorescence in pot-grown plants using an single reflex lens (SRL) digital camera. The assay is highly quantitative and reasonably sensitive when RAW files are quantified with NIH ImageJ software containing the DCRaw plug-in.
In this workshop, participants will photograph GFP expressing plants with a SRL CMOS camera and analyze digital data using NIH ImageJ. Participants will receive plans to build the inexpensive (~$250) epifluorescent attachment, copies of an inquiry investigation performed by General Biology Laboratory students at the University of Detroit Mercy, and instructions on how to download and use the free NIH software. Participants are encouraged to bring USB flash drives (a.k.a., memory sticks).
